دراسة العوامل المؤثرة على التمحفظ الدقيق لليورياز كبروتين نموذج بطريقة التصميم المسحي
Abstract
تهدف هذه الدراسة إلى تحضير جسيمات دقيقة حاوية على بروتين نموذج (يورياز) باستخدام طريقة تبخر المحل عبر الاستحلاب المضاعف م1/ز/م2, ونظراً للعوامل الكثيرة (عوامل الصياغة وعوامل التحضير) المؤثرة على خصائص الجسيمات تم استخدام طريقة التصميم المسحي Screening design لتحديد العوامل المؤثرة رياضياً. درست فعالية التمحفظ (EE) وفعالية اليورياز الحيوية (UA) كخصائص للجسيمات الدقيقة المحضرة. بين التحليل الرياضي والإحصائي أن فعالية التمحفظ تزداد بزيادة حجم المحل العضوي وزيادة سرعة التحريك خلال الاستحلاب الأولي وبنقصان حجم الطور المائي البروتيني. من جهة أخرى انخفضت فعالية اليورياز الحيوية بزيادة حجم الطور المائي الداخلي البروتيني. بالنظر إلى كل العوامل المدروسة (المؤثرة أو لا) تم تحضير جسيمات دقيقة حاوية على اليورياز بفعالية تمحفظ عالية (≈82%) وبفعالية حيوية لليورياز تصل إلى 55% تقريباً وهاتان القيمتان هما الأفضل من بين كل النتائج التي تم الحصول عليها. كان الموديل الرياضي ملائماً للدراسة نظراً لقيمة معامل التحديد المرتفعة: =R2 0.996 و 0,995 لفعالية التمحفظ وفعالية اليورياز الحيوية على الترتيب.
The objective of this study was to prepare microparticles containing a model protein (urease) by double emulsion-solvent evaporation w1/o/w2. A screening design was used to evaluate the effect of the process and formulation factors on microparticles properties.
The microparticles properties considered were the encapsulation efficiency (EE) and the urease activity (UA). The mathematical and statistical analysis showed that the encapsulation efficiency increased with increasing organic phase and stirring speed during the second emulsification. It also decreased when inner aqueous phase was increased. Urease activity was decreased with an increase in the volume of the inner aqueous phase. Following the screening study and the whole results it was possible to prepare microparticles in optimal conditions with high encapsulation efficiency (≈82%) and relatively high urease activity (≈55%). The mathematical model was suitable for the study regarding the high value of R2 (0.996 and 0.995) for EE and UA respectively.
